摘要:本发明公开了一种Ⅰ型、新型鸭肝炎二价卵黄抗体的制备方法及其产品和应用。本发明的一种Ⅰ型、新型鸭肝炎二价卵黄抗体的制备方法,包括收集高免蛋、高免蛋消毒、海藻酸钠除脂、硫酸铵沉淀、抗体的超滤膜浓缩和纯化以及灭活除菌等步骤。研究结果表明,本发明采用海藻酸钠除脂、硫酸铵沉淀提取得到的Ⅰ型和新型鸭病毒性肝炎二价蛋黄抗体,抗体纯度高、效价高、副反应小、治疗效果明显,能够安全方便的用于Ⅰ型以及新型鸭肝炎病毒的同时防治。

主权项:一种卵黄抗体(egg yolk antibodies,IgY)的提取方法,其特征在于包括以下步骤:(1)收集高免蛋;(2)将高免蛋浸入新洁尔灭水溶液中消毒,然后用甲醛熏蒸高免蛋,打破蛋壳,除去蛋清、胚盘和系带,收集卵黄;(3)除脂量取卵黄体积,加入灭菌玻璃瓶内,按其体积的1.5‑2.5倍加入海藻酸钠溶液,混合并调至pH5.0‑5.2,置2~8℃保存过夜;其中,所述的海藻酸钠溶液中,按重量百分比计,含有0.1‑0.15%的海藻酸钠以及0.02‑0.08%的NaC1,其余为纯化水;(4)盐析次日,吸取上清液,调pH7.2,加入硫酸铵至终浓度为28%,再搅拌均匀,离心,弃掉上清,沉淀用PBS缓冲液溶解沉淀;(5)抗体的纯化沉淀溶解液经截留分子量为50KD的超滤膜包过滤浓缩至原体积1/5,加入PBS缓冲液恢复至原体积再进行浓缩,反复3次,获得卵黄抗体原液,置2~8℃保存;(6)抗体的灭活除菌获得的卵黄抗体原液用灭菌生理盐水稀释后经甲醛溶液灭活,然后经滤膜过滤得到最终的卵黄抗体提取液。
