摘要:本发明公开了一种甘肃贝母试管小鳞茎培育的方法,以解决现有贝母培养方法存在的诱导率低、培养周期长、步骤繁琐的问题。它包括以下步骤:选择外植体将贝母掰成鳞瓣和鳞心,对外植体消毒,然后对外植体进行诱导,具体诱导方法是,将灭菌后的外植体接种于诱导培养基中,在温度为16-25℃,黑暗条件下培养,在鳞心或鳞瓣的基部诱导出原球茎;对于鳞心,诱导培养基包括如下配方:1/2MS基本培养液、蔗糖50-80g/L、活性炭2-5g/L、琼脂5-6g/L、精氨酸500-2000mg/L、玉米素(ZT)1-5mg/L、油菜素内酯(BR)0.1-0.5mg/L、茉莉酸甲酯(MEJA)1-5mg/L;对于鳞瓣,激素配方不同于鳞心,培养基的pH值为5.5-6.5。本发明方法具有繁殖速度快、培养周期短、扩繁率高、方便移栽、既可作种鳞茎栽培、又可作商品使用的特点。
