摘要:本发明提供的一种与伊犁马乳用性状相关的SNP标记及其检测方法,该SNP标记为位于马21号染色体上第29764513位的碱基,此处碱基为G的伊犁马日均产奶量显著高于此处碱基为A的伊犁马,利用分子标记技术,检测待测伊犁马的基因型,根据基因型进行伊犁马产奶量的选育,从而加速新疆专门化乳用马品种的培育。

主权项:一种与伊犁马乳用性状相关的SNP标记及其检测方法,其特征在于:1)提取待测伊犁马的基因组DNA;根据GenBank登录号为BIEC2‑560774的序列,设计合成引物,上游引物为5'‑ GGTTGCTATTCTGGGCTTTG‑3',下游引物为5'‑TGTGTTGGGATGGACAAAAA‑3' ;扩增片段大小为194bp,最适退火温度为57.1℃;2)以待测伊犁马的基因组DNA为模板,对设计引物进行PCR扩增;反应体系:PCR Mixture 10.0μl,上下游引物各0.4μl,DNA模板1.5μl,加水至总体积20μl;反应条件:94℃预变性5min,94℃变性30s、57.1℃ 30s、72℃延伸1min,循环30次,72℃延伸5min,4℃保存;3)对扩增的PCR产物,进行SSCP检测;其中电泳的电压180 V,电流50 mA,时间10 h;4)选取个体的 PCR产物,送往上海生物工程技术有限责任公司进行DNA测序,判定位于马21号染色体上第29764513位的碱基G的伊犁马日均产奶量显著高于碱基为A的伊犁马日均产奶量。
