摘要:本发明提供一种快速检测海洋微藻脂肪酸含量的方法,该方法是利用饱和KOH-CH3OH对微藻中脂类提取并造化,使提取过程和造化过程合二为一,再用HCl-CH<sub>3</sub>OH进行甲酯化,获得脂肪酸,该组分经过提取、皂化和甲酯化后进行气相色谱质谱分析,具体步骤如下:(1)样品准备,(2)脂肪酸的提取、皂化和甲酯化,(3)脂肪酸的气相色谱检测条件。该方法具有操作步骤简单,节省时间,方法简单快捷,数据直观,增加了样品之间的可比性。

主权项:1. 一种快速检测海洋微藻脂肪酸含量的方法,其特征在于:利用饱和KOH-CH3OH对微藻中脂类提取并造化,使提取过程和造化过程合二为一,再用HCl-CH3OH进行甲酯化,获得脂肪酸,该组分经过提取、皂化和甲酯化后进行气相色谱质谱分析,具体步骤如下:(1)样品准备:称取8-10mg的藻粉样品;配制饱和KOH—CH3OH溶液和1moL/L的HCl—CH3OH溶液(使pH<2);取15.00mg十九碳酸于10mL容量瓶中,用正己烷稀释至刻度配制成内标溶液;将混合标准溶液用正己烷稀释至100倍成标准溶液;(2)脂肪酸的提取、皂化和甲酯化:将藻粉样品置于螺口试管中→加50μL浓度为1mg/mL的十九碳酸内标和1mL的饱和KOH—CH3OH溶液→充氨气保护→于75℃水浴皂化10min→冷却后加2mL的1moL/L的HCl—CH3OH溶液(使pH<2)混匀→75℃水浴10min→冷却→加0.5mL正己烷和2mL蒸馏水→振荡→静置→取正己烷层用气相色谱质谱仪分析;(3)脂肪酸的气相色谱检测条件:a、色谱柱:HP-5MS石英毛细管柱(30m×0.25mm id×0.25um);b、载气:氦气,流速为1mL/min,进样口温度250℃,传输线温度为300℃;c、电子能量70eV,离子源温度230℃,四极杆温度150℃’;d、升温程序为:初温50℃,10℃/min升到280℃;e、进样量1μL,分流进样(10∶1),扫描方式为全扫描(Scan)方式;各脂肪酸组分含量由对应的峰面积和内标物的峰面积的比值求得。
